- Double digests of M210 to M221 with EcoRI and PstI, run on Gel 77. Results:
- M210-M212 (glgC triple mutant-pSB1A2): two clear bands - one is for the vector (about 2 kb), the other is for the gene (about 1.5 kb).
- M214 (pSB1A2-cenA) generated no bands at all.
- M215-M216 (bglX-pSB1A2): M215 showed two bands, one for the vector, the other for the bglX gene (about 1.5 kb) - this looks promising and should be sequenced. M216 does not contain a band for bglX.
- M217 to M221 (dxs+crtEBI): no bands for dxs+crtEBI (about 5.2 kb), I will re-run the miniprep products on gel tomorrow. (YAN)
- Double digests of M205 to M209 with EcoRI and PstI, run on Gel 78. Results:
- M205 to M206 (SOB2+ISO2): M205 generates a band for ISO2 (2.3kb) but not the SOB2 vector. M206 generates a band for SOB2 but not ISO2.
- M207 to M209 (SOB2+rbs+ISO2): All generated a band for ISO2 (2.3kb) but not SOB2. (YAN)
