  • Performed analytical digests of X17-20 (pSB1A2+dxs+lims) with a) EcoRI and b) EcoRI/PstI. These were run on gel 61, which shows that X20 (streak 5) corresponds to M137 and is pSB1A2+dxs+LIMS. (X17-19 are not the desired product and have been discarded.) (AH)
  • Made X21, maxiprep of pSB1A2+dxs+crtE from plate 130, streak 3. (AH)
  • Made minipreps M160-M181:
    • M160-M163: plate 155 streaks 1-4 (L48, pSB1A2+dxs+lims+appY).
    • M164: plate 155 streak 5 (L49, pSB1A2+crtB+crtI+appY).
    • M165-M168: plate 158 streaks 1-4 (L50, SOB+glgC-mut1,2).
    • M169-M172: plate 159 streaks 1-4 (L51, SOB+glgC-mut1,2,3).
    • M173: plate 160 streak 1 (L55 (SOB+rbs+glgC-mut1,2,3).
    • M174-M175: plate 160 streaks 2 and 3 (L52, SOB+glgC-mut1,2,3).
    • M176-M178: plate 161 streaks 1-3 (L53, SOB+rbs+glgC-mut1,2).
    • M179-M181: plate 163 streaks 1-3 (L56, pSB1A2+PcstA+lacZ). (AH, Yan)
  • EcoRI/PstI digests of M160-M181 were set up ready for running on a gel tomorrow. (Yan)
  • M160-M174 were run on Gel 62: M160-M163 (pSB1A2+dxs+lims+appy); M164 (pSB1A2+crtB+crtI+appY); M165-M168 (SOB+glgC-mut1,2); M169-M172 (SOB+glgC-mut1,2,3); M173 (SOB+rbs+glgC-mut-1,2,3); M174 (SOB+glgC-mut1,2,3). Note: M175-M181 will be run on gel tomorrow on Gel 63. (YAN)
  • PCR of SU1 and ISO2 with glycerol and annealing temp of 60C: P88 (SU1) and P89 (ISO2) (YAN)
