- Minipreps M194-M197: Appy+dxs+Lims (YAN).
- Double digests of M182 to M193 run on Gel 68:
- M182 to M187: pSB1A2+cenA
- M188 to M193: crtBI+appY
- The bands were not very clear; however, there were bands for crtBI+appY at 3 kb position, and bands for cenA+pSB1A2 at the 2.3 kb position.
- Re-run M182 to M193 on Gel 70.
- The gel did not look good! The bands for cenA did not exist, nor did the bands for crtBI+appY, so the ligation of appY into crtBI failed! (YAN)
- Run PCR products P91 to P94 on Gel 69.
- P91 to P94: SOB2-SU1, SOB2-rbs-SU1, SOB2-ISO2, SOB2-rbs-ISO2.
- The gel looked good! (YAN)
