- Made 17 minipreps from colonies on plates 83-86: M80-83 from plate 83 (L26: pSB1A2+rbs+appY), M84-85 from plate 84 (L27: pSB1A2+rbs+crtB), M86-90 from plate 85 (L28: pSB1A2+cenA) and M91-96 from plate 86 (L29: pSB1A2+cex). This was followed by double digestion of all the miniprep products (M80-96) with EcoRI/PstI. (Yan, OG)
- Third glgC mutagenesis performed by MABEL on BABEL2+glgC-mut1,2 (since pSB1A2+glgC-mut1,2 appears to have failed): P50 (Yan)
- P50 and double digestions of M80-M96 were run on gel 36. Results were good for M80-82 (pSB1A2+rbs+appY), M84-85 (pSB1A2+rbs+crtB), M92 (pSB1A2+cex) and P50 (glgC-mut1,2,3). Other results were poor, with bands in the wrong position for M86-M90 (pSB1A2+cenA). (Yan, OG)
- Sequencing results back today indicate that M50 is pSB1A2+rbs+crtI, M63 is pSB1A2+rbs+crtE and M67 is pSB1A2+rbs+crtB. (AH)
