- P95: cex mutagenesis with M151 as template. Annealing temp 58 degrees, extension time 68 seconds. (Note: there was another microtube in the PCR product box, labelled as "cex mut", and I did not know who did it. So, I repeated this mutagenesis again, PCR products labelled as P95) (YAN).
- L62: ligation of dxs+crtE (digested out of M155) and crtBI (digested out of M110). Note: crtBI as vector, cut with EcoRI and XbaI; dxs+crtE as insert, cut with EcoRI and SpeI (YAN)
- Transformation of L58 to L61, followed by sub-cloning and overnight culturing (C.F)
