Wet Lab

B.subtilis

• The transformation protocol 1, click here for link to the protocol, was checked today. The plate containing B.subtilis transformed with no DNA had no colonies, B.subtilis transformed with 40ng DNA contained ~10 colonies and B.subtilis transformed with 400ng DNA contained ~10 colonies.

Cloning

• Double digest results from the 21st checked. A second gel was visualised early (but no image recorded) to allow identification of the small Termiantor insert
• Insert was succesfully identified on the gel
  • All Registry sequences that were succesfully cloned appear to be ok for use
