September 15th
Transformation
The DNA ligation over the weekend has been transformed into TOP10 made chemically competent.
Induction
To test for R0080 function, we're trying to visualize luciferase expression on an SDS-PAGE. Therefore we made a 1.33 M arabinose stock (1000x) for induction [1] (page 3) and have grown K115012 and K115011 o/n both with and without induction.
Colony PCR E. coli genes
A colony PCR on eight colonies for atoB, idi and ispA were performed. The PCR was performed o/n, after the program the machine was programmed to keep a constant temperature of 4 degrees Celsius. The result will be checked on gel tomorrow.
