September 24th
Colony PCR
The transformations of K115022 and K115027 showed plenty of transformants. We've done a colony PCR on them and they gave good results. We'll work further with the most left of both the different cultures on Monday for prepping and further construction of K115030 and K115035.
Luciferase assay
We've lysed all cultures (30ºC and 37ºC) for measurements now, both were around OD=1. Measurements could not be done today due to unavailability of the hardware, therefore we've also started 42ºC cultures, and an extra 30 and 37ºC cultures of part K115036, as this part was proven well transformed yesterday. Tomorrow we should be able to do measurements.
Primer solutions
New primers were recieved from Invitrogen. Stock solutions (100 uM) and working solutions (20 uM) were made and will be used tomorrow in TouchDown-PCR.
