August 27th
Colony PCR
The Colony PCR on colonies transformed with punches showed incorrect products, indicating the transformations went wrong (it looked like contaminations). We did find out pUC18 gives product as well.
Competent Cells
We made 50 eppendorf tubes that contain 100ul worth of competent cells each. We expect this to last the rest of the project. Again, the protocol of OpenWetWare was used. The cells still have to be tested with a positive control.
