A colony PCR was done on the transformed cells. The gels are depicted in figure 1. AtoB, idi and ispA are listed in the TUDelft registry as K115050, K115056 and K115057 respectively. Sizes of the parts + biobrick primer DNA should be:

  • atoB: 1504
  • idi: 868
  • ispA: 1219
  • K115030: 1604
  • K115032: 1648
  • K115033: 1604
  • K115035: 1635

K115030 and K115035 seem properly transformed. K115032 didn't seem any better as it was in the previous construction. We'll just keep using that plasmid. K115033 didn't look like it was a succes.

Restriction and Ligation

3A assembly of K115033 and normal assembly of atoB using XbaI and PstI for restriction were performed. Ligation was performed o/n @ 16ÂșC.
