Wetlab work
Checked on plates in SC incubator
- Grace
- Checked on cell density and triparental conjugation plates
- No cyanobacteria growth observed yet (they like to take their time). No contaminants observed.
Alkaline lysis prep
- Grace & Krystle
- Inoculated 5 ml LB+sp50 with 1 colony DH5α transformed w/ pRL1383a. Incubated overnight at 37C.
Restreaked DH5α w/ pRL1383a plate
- Krystle
- Restreaked on LB+sp100. Incubated at 37C overnight.
Drylab Work
Completed Biosafety Training

- Edited primers for nir promoter
Alkaline lysis

- Researched protocols for alkaline lysis plasmid prep. Plan on doing plasmid prep tomorrow.
