Plasmid prep

- Plasmid prep of E0240, J32007, I14032 ('05 and '08 distributions), I51020
- Centrifuged for 90 min. at 4C after adding KoAc
Cryostock

- Did not take out 2.4 ml of E. coli before doing plasmid prep (oops). Reinoculated for cryostocking tomorrow.
Construction of p+r+g, g+t, J33207+t
- Grace
- RE digest of nir+rbs, plac+rbs, GFP, B0015, J33207
- Digested nir+rbs, plac+rbs, J33207 with EcoRI and SpeI
- Digested GFP, B0015 with Pst1 and XbaI
- Ran digests on EtBr stained 2% agaorse gel
- Gel did not run despite using new TAE buffer
- Redid RE digest overnight
Made KoAc solution for plasmid preps

- Ran the large-scale PCR of aadA(BB), aadA(pRL1383a), and P1 lytic region on a gel, cut it out, and purified it using the Qiagen Kit
