- Prepare low salt LB plates w/ 50ug/ml BLAST
- Prepare blast stock solution, 50mg + 1ml
- make new LB plates
- make YPD plates
- prepare ligation mix using T4 Dna Ligase and LiP and pPIC6
- spread x-33 yeast cells onto ypd plates, place in 28C to incubate
