- Repeat digestion protocol using NotI, EcoRI, LiP, and pPIC6
- prepare and run 1% agarose gel, cut out products
- perform gel purification using QIAgen kit
- prepare and run 1% agarose gel
- grow x-33 yeast in YPD medium overnight
- make ligation mix with T4, Lip, and pPIC6
- grow overnight culture of xl1blue
- do transformation using ligation mix, spread onto plates, incubate overnight
