Team:Mississippi State/Transformation of pPIC6alpha into E.Coli

  • The vector(We used pPIC6alpha A) comes in dry form, 20ug.
  • We added 20ul ddH2O to get stock solution of 1ug/ul of the vector.
  • Then, slow vortex followed by short spin.
  1. Add 100ul competent cell solution to 1.5ml tube.
  2. Add 0.2ul pPIC6alpha stock solution to cell stock in tube.
  3. Incubate on ice for 30min.
  4. Incubate in H2O @ 42C for 2min. (Heat Shock)
  5. Immediately add 500ul LB.
  6. Incubate at 37C with agitation for 1hr.
  7. Spread cells to 3 plates.
  8. Incubate at 37C for overnight(14-16hrs).
