The Day of Strange Results

  • The Purple Russian transformants did not grow yet again. This time it might be because Jason made a mistake while transforming the competent cells. We should probably just start from the beginning and try to PRC a new Biobrick and transform it.
  • The strange result from the Colony PCR on the Tryp brick is still a mystery. It could be because the biobrick might have an internal Xba or Pst site (remember, we didnt optimize these bricks or have them synthesized) that got cut when we did the digest. We should check the sequences and have the mystery insert sequenced.
  • Tom transformed B0043 yesterday. He wasnt sure if the part had Amp resistance or Kan resistance so he plated the transformants out on both to be sure. Both grew. Each brick should have only one resistance gene so this is very strange indeed.

Not so Strange Things

Crude and Soluable Proteins of 25, 35 and Thiolase in I0500
  • Winnie ran the sequencing she did yesterday on the Blue Ox brick up to the MBSU. Should get the results back tomorrow sometime.
  • Jason is doing PCR to try and remake all three reporter biobricks.
  • Did Western transfers of crude and soluable protein of 25, 35 and thiolase in the I0500 vector (see image at right). Next step is the antibody detection.
  • The night team transformed the Purple Russian, the Blue Ox and the Tryp plasmids into E. coli and set the plates to incubate overnight.
