[x] reinoculate pJET + gene 6
[] inoculate 2 tubes each of a couple RBS-gene6 samples that looked good based on Friday's colony PCR results
- Monday: make glycerol stock from one tube of each, and miniprep the other tube of each for diagnostic digest* and gel)
Diagnostic digest* (Monday)
Do 2 diagnostic digests for each sample: Do insert excision using a biobrick enzyme double digest like normal (to check for correct insert size), but also use ApE to see if there's an enzyme that Charles lab has which will cut either a) RBS (preferable) or b) gene 6 (it can cut it once or twice, as long as the band sizes will be large enough to see on a gel). Do a separate single digest with this enzyme (hopefully there is one you can use) to check for linearization of the plasmid.
