SENSING
1) LuxS test with YFP works, but not in standard form due to a standard restriction site in the part.
2) Fused receptor construct (Tar/LuxQ) exists, but not in standard form, standardization is not possible anymore due to lack of time, chemotaxis and kinase assays did not show significant functionality

PHAGES
1) Chloramphenicol and CI in standard form, only minimal characterization possible and done
2) OriT cloned into standard form and sequenced, same part as already existing in Registry (according to sequence information), characterization is in progress
Plan: We will submit oriT as a new part, since we generated new and reliable, physical DNA. All sequencing results will be provided.

COLICIN
1) Sender (LuxI under constitutive promoter) as composite part in standard form, characterized in top10 by calibration of supernatant with commerical AHL (AI-1), read-out is induction of LuxR-regulated GFP. Result: Synthesis rate k for AHL depending on colony forming units (cfu) or optical density. Prey strains (top10) with Sender and GFP genes are available.
2) ColE1 cassette is a composite part in standard form. Works. characterization in progress with different AHL, killer, and prey concentrations. Characterization by visualization with microscopy to be done. Chemotactic strain with ColE1 available, but not yet with addtional plasmid for mcherry.
3) ColE9 short cassette is a composite part in standard form. works. not characterized.
4) ColE9 long cassette is a composite part in standard form. works. not characterized.
5) Toxicity test of ColE1 with MCF7 cells planned with microscopy (Yara)

IMPORTANT
We will provide a summary of all deliverables plus a list of all constructs to clearly point out to iGEM their status, when we send them there.

WIKI and PATENT
The patent will probably filed next Monday. WIKI deadline is next Wednesday, and it might happen that the servers will be overloaded. Try to hold back the sensible information until Monday! If not possible, do not wait until it is too late! Kolja or me will ask for password protection.

OPEN DAY
Next Monday starting at 9 am we will have an open day. A general introduction several experimental stations are planned with instructors explaining what is going on. Pictures will be taken and a PhD will be at the microscopes.

TRANSPORTATION
departure by train will be from Heidelberg main station at 9:15 am. Philipp and Dominik will be picked up by Roland
Chenchen will organize the transfer from Boston airport to Cambridge hotel.
Cambridge hotel to Stata center is in walking distance (15 min).

ACCOMMODATION
We extended the number of rooms. There is enough space that two post-docs can share one room. A sign-up sheet is available on the internal wiki under NOTES.
