Sequence gel purified fragments from yesterday
Transform I0500
Digest J61003 with Xba/Pst to insert ORF's
Digest minipreps with Xba/Pst to insert into J61003
Run gel of digests and excise correct fragments
Ligate vector/inserts
Transform ligation reactions
Lab clean up

Jason:

  • Started investigating the Cry1 blue light detection pathway
  • Started to build the LUX operon plant biobrick
  • Went to Dougs awesome Modeling Course

James:

  • Checked all O/N's set up by SZ and WX - all grew except 023 WX. Will set up again tonight.
  • Followed standard Qiagen miniprep protocol for all o/n's.

Winnie and Saima:

  • Digested J61003 using Xba and Pst
  • Digested minipreps of last year's genes using Xba and Pst
  • Began sequencing the o/n minipreps

In 2004 Texas submitted the cyanbacterial equivalent of our system, so... Our biobricks would be unique but they would do the exact same thing.

Lab Tip of the Day

When using the NanoVu to measure your DNA concentration, please ensure that you press the measure button and not the calibration button. Also, remember that the instructions for using the NanoVu are posted above the NanoVu. Please refer to these instructions in times of doubt instead of getting Chris to do it for you.
