lab work (JD): 

             Counted colonies on both mine and Shannon's plates from yesterday's ligations
                              -unfortunately equal numbers on negative plates and ligation plates so ligation 
                               did not work
                              -my plate had 100s on the negative control so I assumed the vector was not cut 
                               properly
             Conclusion: Re-digest K102011 with more enzyme, for 2 hours and run very slowly on gel
