lab work (JD): 

             From plates of yesterday's ligations there were NO colonies
             Replate cells on bottom of tubes on LB+kan35 again
             Carried out calculations to better determine appropriate dilutions and protocol for annealing, 
             digestion and ligation of newly created oligos.
