  • Pipette 10mL LB into red-capped culture tubes or conical shake flasks.
  • Using an autoclaved toothpick, gently touch the point to a single colony.
  • Drop the toothpick point down into the LB.
  • Incubate overnight at 37°C whilst shaking.
