Tuesday 26th August
- Due to unforeseen circumstances, restriction mixtures were put in the fridge on Friday 22.08.08. Therefore the restriction was allowed to run for a further 1.5 hours before running on a gel.
Lane 1: 1kb ladder
Lane 2: empty
Lane 3: pUC57-ncl08 restricted with EcoRI + NHeI
Lane 4: pUC57-ncl08 restricted with NHeI and BglII
Lane 5: pUC57-ncl08 buffer A control
Lane 6: pUC57-ncl08 buffer M control
Lane 7: pure pJWV021
Results from overnight (ON) agar cultures:
- pGFPrrnB (amp) - no colonies
- pGFPrrnB (spec) - colonies present
- pGFPrrnB (no antibiotic) - colonies present
- ON cultures were made:
- pGFP-rrnB grown on ampicillin
- pJWV021 grown on ampicillin
- pUC57-ncl08 grown on ampicillin
