PCC 6803 Transformation

  1. 5ml cells in BG-11 (OD730 = 0.8), grown in BG-11 + 2 mM EDTA for 2 days prior to transformation to increase transformation efficiency
  2. centrifuge @ 6000g fo 15 min at room temp.
  3. resuspend pellet in 0.5 mL BG-11 (~109 cells/mL)
  4. add plasmid DNA to Cf = 10 µg/mL (Cmin = 0.02 µg/mL)
  5. incubate for 5hrs at 30°C in light (50µmol/photos/m2/s & cover in tissue paper?)
  6. grow in BG-11 w/o antibiotic for 1 day (optional)
  7. plate onto antibiotic containing plate
