Making PCR Gel

  • the Gel is a 1% Agarose gel
  • 0.25g/25ml Agarose/TAE
  1. Weigh 0.25g Agarose into flask(Remember weight).
  2. Add 25ml TAE.
  3. Microwave 1min to boil
  4. wait 7min
  5. Reweigh
  6. Add H2O to difference.
  7. Add 5ul EtBr (Wear Gloves)
  8. Pour into gel box, inserting templates.
  9. When gel dries, remove templates and pour TAE until it reaches top of box
  10. Make sure nodes are on back side
  11. get ladder (1ul)
  12. touch to TAE first, then load first well
  13. get 1ul loading dye, place drop on parafilm
  14. put 2ul PCR product on drop and mix.
  15. load 2nd well
  16. put PCR product on ice(-20C)
  17. Hook up gel, black-left, red-right; where black is (-) and red is (+).
  18. set to 80V for 60min.
  19. Remove gel and take to get picture.
