Overnight Results

  • pSB1A10 screening plasmid grew just fine
  • Assembly 1 (BP1 + Term) grew more colonies
  • pSB1AC3 and pSB1AK3 both grew a small number of colonies


Goals

  • Transform overnight ligations
    • Note: Considering the other ligations didn't work and the restriction digest might be the problem we don't expect much
  • Make small (10ml) broth of plates with colonies
    • pSB1AC3, pSB1AK3
  • Repeat assemblies
