pMPM-T5+AID+T7 (AID and T7 transcriptional fusion)

Based on: pMPM-T5-AID.
Resistance: tetracycline
Features: Transcriptional fusion of AID protein and T7 polymerase (proteins transcribed together but translated independently) under control of arabinose promoter.
Preparation: T7 polymerase PCR amplified ( T7lRBSHi T7pXbSal primers) from Rosetta genomic DNA and digested with HindIII and SalI. Digested fragment was cloned into pMPM-T5-AID [notebook entries]
Usage: Transcriptional fusion of AID and T7 polymerase. AID should change sequnces independently of T7 polymerase. Sequences under T7 promoter should be highly transcribed and have higher AID-induced mutation rate.
Vector was used in blue/white and rifampicin tests.
