Using the Nanodrop Spectrophotometer, the concentration of the pWW53 plasmid was found to be approximately 300 ng/uL.  We are going to check the concentration one more time tomorrow, to verify the concentration of the DNA.

We also streaked out colonies from the overnights on LB-Agar + 10% Indol to see if we could get blue colonies.

The overnights did not grow as well as expected, but we did see some growth. We decided to carry out a miniprep to attempt to isolate the plasmid.

Using the GenElute™ Plasmid Miniprep Kit from SIGMA-ALDRICH, we isolated the pWW53 plasmid from Pseudomonas putida. For elution step 9, GIBCO Molecular Biology Reagent Water was used instead of the Elution solution from the kit.

Using the Nanodrop Spectrophotometer, the concentration of the pWW53 plasmid was found to be approximately 300 ng/uL. We are going to check the concentration one more time tomorrow, to verify the concentration of the DNA.

We also streaked out colonies from the overnights on LB-Agar + 10% Indol to see if we could get blue colonies.
