Using the transformation completed by Wayne (12/06/07) of XL1-B cells with I732018 and the overnight cultures, DNA was isolated by QIAprep standard miniprep protocol. I732018 was sequenced. I732018 was digested with XbaI + PstI. No evidence of DNA in the gel. Retry!
lab work (JD):
Quantitated K102005/BamH1 and Nco1 Ligated K102005/BamH1 and Nco1 with TA0/BamH1 and Nco1 (1/100 dilution) to create plasmid K102007 Transformed XL1-B cells with K102007 and negative control Plated cells and left them for O/N
