lab work (SD): 

            Isolated DNA (K102002) from LB culture tubes from 18/06/08 using QIAprep spin miniprep protocol.
            Set up for sequencing.

lab work (JD): 

            Counted and recorded number of colonies on newly plated K102007 plates from yesterday
                    K102007 with the newly recut TA0: 128
                    K102007 with heated old TA0: 162
                    Negative control: 176
             hence ligation didn't work AGAIN, tomorrow we will try something different and redo ligation on Monday
             Mini-prep of 7 O/N colonies of K102003.1-K102003.7
             Set up for sequencing
