Today
Chris
- Ligated Tryp into J6. Since the result of the ligase test from yesterday was inconclusive (because we didnt run any controls!) this may not work.
Saima
- Digested LacIERE with XbaI/Pst. Ran the digest on 2% gel. Digest didn't work!
- PCRed LacIERE Miniprep. Ran the PCR product on 1% gel. Unfortunately, gel was tossed out. Need to redo this.
- Colony PCRed Cam35s+J6 Ligation to double check if it is what's expected. The band sizes were totally off.
- Transformed Tryp+J6 Ligation.
- Transformed revised parts that just came in: Revised ER01, ER02, BisDB and BisDA.
