Yeast Shuttle Plasmid
- A plate of yeast strain BY4741 (MATalpha, ΔBar1::Kan MX, his3Δ1; leu2Δ0; met15Δ0; ura3Δ0) was obtained.
- The yeast was incubated overnight at 30 degrees Celsius.
LuxR under AraC and TetR
-Obtained BBa_R0040 (TetR), BBa_R0080 (AraC)
-transformed and plated with Amp
LuxR under pLac
-Obtained parts R0010, I0462, and J04430.
-R0010 and I0462 were transformed into MG strains, and plated on Amp.
-J04430 was transformed into DH strain, and plated on Amp.
