Team Sorbitol:

Digested the srlD and pET28a with BamHI and HindIII
Extracted the digestion and performed a ligation which ran overnight at 4C
Another attempt to PCR srl Operon success! 7 of 8 reactions correct band placement on gel
Digestion and Ligations performed. Success!
Team Fungus:
Made 1 : 10 dilutions of C and N-limited cDNA in 10mM Tris-CL (pH 8.0) to preserve the little we have left.
Ran gradient PCR with Taq with diluted cDNA.
Ran nested PCR with Taq with diluted cDNA.
Ran nested touchdown PCR with Taq with diluted cDNA.
