BioBrick Promoter Measurements
- Thawed glyerol stocks for TOP10 cells containing promoter constructs I20260, I20268, I20269, and I20270.
- Inoculated cultures on four different agar plates containing LB and kanamycin to prepare for measurements.
LuxR from AraC and TetR
- Selected five colonies from old AraC plate to grow overnight: 3 ml Tsy + 3 ul Amp.
- Plated AraC from glycerol stock in Agar LB + Amp plate.
- Resuspended four primers in deionized water to the concentration 100 pmole/ul, later stored in -20 Celsius.
LuxR from pLac
- Started overnight culture of I736004 for sequencing and glycerol stock.
- Started overnight culture of R0010 for glycerol stock.
