-Picked the colonies which appear to have amplified during the screening PCR, and Incubated them in LB+amp media at 37.0C overnight.

-Plasmid Prepped the colonies which were incubated overnight. Ran Plasmids on a 1% Agarose Gel at 100V for 27 minutes.

-Visulaized the Gel, and chose appropriate cells to glycerol stock.

-Performed a Restriction Digest on RS1 and RS2 from pGEM T-easy using XbaI and SpeI. Ran overnight at 37.0C

-Reran the PCR of the riboswtich from the primers and has been working on the Riboswitch throughout the month.
