| 1. Transform Dual Promoters: transform the dual promoters received from GeneArt into DH5alpha competent cells. The Dual Promoters are (1) LacI and LAMBDAcI, (2) TetR and P22mnt. Follow transformation instructions in beginning of wiki. Place dual promoter DH5alpha cells in 1mL of 2xTy broth and incubate @ 37C with mixing @ 220rpm's for 2 hours to allow cell growth. |
