- Plasmid resuspension from IGEM paper spots
- Transformation
- Plasmid extraction
- BioBrick digestion with restriction enzymes
- DNA gel extraction
- Antarctic Phosphatase
- Ligation
- PCR
DNA gel extraction
(Estimated time: 1 hour and 30 min)
Materials needed:
- Agarose gel with well-separated, sharp bands
- Scalpel and tweezers to cut gel (they have to be cleaned in ddH2O after every cut)
- Roche Agarose Gel DNA Extraction Kit
