Today

Chris

  • Ligated Tryp into J6. Since the result of the ligase test from yesterday was inconclusive (because we didnt run any controls!) this may not work.

Saima

  • Digested LacIERE with XbaI/Pst. Ran the digest on 2% gel. Digest didn't work!
    • PCRed LacIERE Miniprep. Ran the PCR product on 1% gel. Unfortunately, gel was tossed out. Need to redo this.
  • Colony PCRed Cam35s+J6 Ligation to double check if it is what's expected. The band sizes were totally off.
  • Transformed Tryp+J6 Ligation.
  • Transformed revised parts that just came in: Revised ER01, ER02, BisDB and BisDA.

Jason

  • sequencing from friday doesn't give comprehesive result
    • will redo sequencing with Vf and Vr and look for proper restriction site.
