lab work (SD): 

            Extracted DNA from gel using QIAquick gel extraction protocol.
            Quantified DNA and ligated K102031 /E+P and K102032 /E+P together.
            Transformed XL1-B cells with the ligation product.

lab work (JD): 

           Ligation of K102011 with TAIn_2, TAIn_3, TAIn_6 and TAIn_7
           Transformed XL1-B cells with the ligations
           Plated on LB+kan35 plates
           Incubated at 37C overnight
