Accomplished

  • Looked at stained gel of 1st order assemblies (47-50). No inserts were visible. This suggests the assemblies are not correct.
  • Bought XbaI
  • Cut out B0034 (RBS) out of gel and ligated it with BP1-3 and E0040 (cut out of previous gel) using higher amounts of DNA than before.
  • Transformed and plated ligations
