Making Overnight Cultures from Frozen Glycerol Cell Stocks

- Pippette 10mL LB into a red-capped culture tube or conical shake flask.
- Using an autoclaved toothpick, take a scraping of the frozen glycerol stock and drop it into the tube/flask. (Alternatively a pipette tip can be used and discarded after transferring the cells to the LB.)
- Incubate at 37°C overnight.
