lab work (SD): 

             Isolated DNA of TA2, TA3, TA6 and Ta7 from overnights using QIAquick spin miniprep standard 
             protocol.
             Sequenced TA2, TA3, TA6 and TA7.
             Made overnights of the three colonies found on the pSB1A3 plate.

lab work (JD): 

             From ligations:
                      TA2In=76 colonies
                      TA3In=90 colonies
                      TA6In=66 colonies
                      TA7In=68 colonies
                      Negative=4 colonies (from Wayne)
               Created 4 O/N from each plate
               Annealed TA2, TA3, TA6, TA7
               Nucleotide removal kit for TA2 TA3 TA6 TA7
               Restriction Digest of TA2, 3, 6, 7 with BamH1 and NcoI
               Ran on gel 2% at 150V for 2 hours
