Team Sorbitol:

Harvested the DNA from these cultures and digested - however they were not the right size so will reperform the ligation
Team Fungus:
Ran PCR purifications.
Ran PCR product out on gel. Strong band!!!!
Run another PCR using Herc II with previous PCR product as template. STRONG BANDS!!!!!!!! Not using Taq is the secret.
