Cloning
- Cloning of BC1 into AB
- Using the simplified ligation protocol.
- Cloning of CD into LuxI+ (LuxI with RBS and Term.)
- Here a modified version of the ligation protocol was used, because the insert is quite big (over 2kb).
- Reaction mix:
- 3μl vector
- 5.5μl insert
- 1μl Ligase Buffer with ATP
- 0.5μl T4 Ligase
=> Cloning worked.
