Site mutation of dxnA and dbfB failed. We suspect that the concentrations of the two fragment are far different so the annealing step has an unsatisfying yield. We perform the same experiment for a second time. And we will make sure the concentration of each fragment.

T7 termitor ligation failed. There are a couple of reasons.The incubation time may be too long.And the ligation effciency may be a bit low.
