Cloning

• Terminator, mRFP - Terminator and pSB1AK3 plasmid containing XL1-Blue cells replica plated and overnight cuture prepared

B.subtilis

• The transformation performed on 15th August 2008 was successful yielding between 1-10 transformed colonies. It was noted that the efficiency of transformation increased with increasing concentrations of DNA. We wish to repeat this protocol, this time using a negative control of competent cells transformed with just water. To do this 20ml of overnight culture of B.subtilis was inoculated.
