WetLab

  • Performed midi preps for Terminator, GFP-T, RFP-T and AK3, all had high concentrations and good yields
Lanes : M = Marker, 1 = GFP-mut3b - terminator Midi 1 Digested, 2 = GFP-mut3b - terminator Midi 1 Undigested, 3 = GFP-mut3b - terminator Midi 2 Digested, 4 = GFP-mut3b - terminator Midi 2 Undigested, 5 = mRFP - terminator Midi 1 Digested, 6 = mRFP - terminator Midi 1 Undigested, 7 = mRFP - terminator Midi 2 Digested, 8 = mRFP - terminator Midi 2 Undigested, 9 = CAT midi 1 Digested , 10 = CAT midi 1 Undigested, 11 = CAT midi 2 Digested, 12 = Catmidi 2 Undigested
  • All insert and vector sizes appear to be correct with minimal contamination
  • Double digests with Spe and EcoRI were performed on GFP-T and RFP-T and a 1% agarose gel run. The AK3 and terminator constructs will be digested and run a gel tomorrow morning.
  • Optimised vector genomics and single colony PCR protocols was also performed today. The results of these will be written out in more detail tomorrows notebook.

DryLab

  • Completed production of synthetic video. Updated wiki with regards to generation of synthetic trajectory and bacteria characteristics used to create synthetic video.
