Let transformed cells on original LB-amp plate grow up at RT. Streaked individual colonies onto 2 plates--one for stock, and one for experiments. They are plated on X-gal/Amp plates. Let sit at RT 6 hours, then 37C for 12 hours. Tomorrow, take out of incubator and dose the experimental strain with UV light for 30s. Put in incubator for an hour to recover (and hopefully change colors). Also plated lacZ stab on X-gal plate. These might turn blue.
