September 2nd

-We carried out the following experiment:
      Four strains in LCCs with SP medium.
      1 % Galactose induction at the beginning. Initial O.D. 0,7
      275 rpm.

-Team meeting: Checking project progress.

September 3rd

-We characterized LCC temperature evolution with water.

September 5th

-We characterized LCC temperature evolution with SP medium and SP medium with bleach (to avoid contamination).

September 8th

- We observed the culture after the experiment under the microcope. There was no contamination but we did observe some elongated microorganisms, same size as S. cerevisiae.

- We discovered that our plaque with the four strains for the following experiment had suffered cross contamination.

September 9th

-We carried out the following experiment:
      Four strains in LCCs with SP medium.
      1 % Galactose induction at the beginning. Initial O.D. 0,6
      270 rpm.
       Initial temperature 31 ºC. Too high.

September 11th

-We repeated the same experiment:
      Four strains in LCCs with SP medium.
      1 % Galactose induction at the beginning. Initial O.D. 0,6
      270 rpm.
       Initial temperature 31 ºC. Too high again.

September 15th

-We repeated the same experiment:
      Four strains in LCCs with SP medium.
      1 % Galactose induction at the beginning. Initial O.D. 0,6
      270 rpm.
       Initial temperature 28 ºC. Perfect!

September 20th

-Trying to reproduce the previous experiment...       Four strains in LCCs with SP medium.
      1 % Galactose induction at the beginning. Initial O.D. 0,6
      270 rpm.
       Initial temperature 28 ºC. But room temperature too high

September 24th

- We repeated the same experiment:
      Four strains in LCCs with SP medium.
      1 % Galactose induction at the beginning. Initial O.D. 0,6
      270 rpm.
       Initial temperature 28 ºC.

September 26th

- Team meeting: Checking project progress and inmediate future actions.

September 27th

- We tryied and increased initial O.D.
      Four strains in LCCs with SP medium.
      1 % Galactose induction at the beginning. Initial O.D. 2
      270 rpm.
       Initial temperature 28 ºC.
