iGem Meeting Minutes 6/10/2008

The meeting began at 3:10

  • Advice from Professors (Rao and Golding):
    • try expressing a T-cell receptor in yeast
    • try using an FC receptor attached to an antibody-allows it to be generic and usable with various antibodies
    • Potential Challenges:
      • Get yeast--ask Dr. Orleans. He is in India for a month but go to his lab. Another option is Dr. Zhao
      • We need a pheremone for the schmoos to come together
        • Can we buy a pheremone? we should be able to, but look into details.
      • We need a reporter
        • GFP or lacZ(colorimetric reaction). lacZ maybe a better choice.
    • To Do this week and early next week:
      • Get yeast and begin culturing--it usually takes 2 days at room temperature in YPD media.
      • Get powdered media and pour it into dishes
      • Order NEB catalog
      • See what the yeast stock center in Seattle, has to offer
        • They should have free fluorescent proteins

4:00 Dr. Rao will take 4 people into his lab and teach them how to clone DNA

  • Dr.Rao explained the procedure for Tuesday's experiment
    • Step 1 amplification-PCR
    • Step 2 separation-gel electrophoresis
    • Step 3 cut plasmid using restriction enzymes
    • Step 4 add gene A by binding sticky ends with DNA ligase

4:40 We decided who was going to be in each group. The groups are posted on the Powerpoint. If you weren't at the meeting pick a group to work with and contact the group members.
